Polydatin Simply Explained
Polydatin is a plant substance that occurs mainly in knotweed species. This page explains in simple words what studies show about it and what remains open.
1. What It Is About
Polydatin is a plant compound. It belongs to the polyphenols, a large group of small plant compounds. Chemically it is closely related to resveratrol, which many know from red wine and grapes. Polydatin additionally carries a sugar unit. This makes it well soluble in water and easier for the body to absorb. It is found in knotweeds such as Polygonum cuspidatum, as well as in grapes, peanuts and red wine. In the plant it acts as a defence substance against pests and against stress caused by oxygen compounds. It is mostly obtained from knotweed roots, sometimes also produced synthetically. What happens in the gut is important for research: gut bacteria convert part of the polydatin into resveratrol. How much of it actually reaches the blood therefore depends on the individual's gut flora. Blood levels therefore do not simply indicate how much has been absorbed. Many human studies also use finely ground preparations to improve absorption of the substance.
2. What Studies Show
Polydatin is best studied in irritable bowel syndrome, a disorder with abdominal pain and irregular bowel movements. In a controlled study with 54 adults with irritable bowel syndrome and 12 healthy controls, participants received a combination of polydatin and the compound palmitoylethanolamide, or PEA, for twelve weeks. The study was randomised, double-blind and placebo-controlled — that is, allocation was by chance, no one knew who received what, and one group received a sham preparation. After twelve weeks, certain immune cells of the intestinal mucosa that fuel inflammation were less numerous. Self-produced messenger substances involved in pain and inflammation also approached normal values 1. However, the studies always examined the combination, never polydatin alone. A second controlled study followed 70 children and adolescents aged 10 to 17 years with irritable bowel syndrome over twelve weeks. Here too, one group of children received the combination of polydatin and PEA, another group a sham preparation. In the treatment group more children became symptom-free, abdominal pain occurred less frequently and bowel movements became more regular. The children tolerated the treatment well 2. The same combination was used for three months in women with chronic pelvic pain. In more than half of the women the pain decreased markedly. There was no sham-preparation comparison group in this study, and long-term data are still outstanding 3. In cell and animal studies polydatin also shows effects on inflammation, on the metabolism of cancer cells and on cells that the coronavirus infects. In an animal model with tongue cancer cells the number of metastases in the lymph nodes decreased sharply 4. In cell studies on SARS-CoV-2, polydatin helped to restore small cell components that the virus disrupts 5. Such findings apply to cells or animals, not to humans. In the controlled study with children no serious adverse effects were reported 2.
3. What Remains Unclear
The most important open point: in the two controlled studies on irritable bowel syndrome described, polydatin was tested only together with PEA. Whether polydatin alone has any effect is therefore not clarified. It is also open whether the combination yields more than the individual substances. For cancer, viral infections and metabolic diseases the observations so far come almost exclusively from cell and animal studies. Whether they apply in humans is a testable question that remains open. How much polydatin is useful also cannot be derived from the available studies: the amounts used varied widely, and studies that systematically search for the appropriate amount are still outstanding. Long-term tolerability and the question of whether polydatin interacts with medicines are likewise unclear.
4. What This Means in Everyday Life
Anyone reading about polydatin quickly encounters large claims: inflammation, cancer, viruses, metabolism. The robust human findings concern mainly irritable bowel syndrome and chronic pelvic pain, and they apply to a combination, not to polydatin alone. The two controlled irritable-bowel studies show improvements in adults and in children, and tolerability was good in these investigations 1 2. For classification this means: polydatin is a well-studied plant compound with initial hints from clinical studies — but not an agent with proven efficacy in the many diseases so far examined only in cells and animals. The next step would be to clarify whether polydatin also works without PEA and what a dedicated controlled study shows.

What is Polydatin?
Polydatin is a plant substance from knotweed plants, grapes, peanuts and red wine. It is closely related to resveratrol, but has an additional sugar unit that makes it more water-soluble. In the intestine, gut bacteria convert part of it into resveratrol.
Does Polydatin Help with Irritable Bowel?
Is Polydatin Well Tolerated?
In the study with children, no serious adverse effects were reported 2. What happens with longer-term use and whether polydatin interacts with medicines remains open.
