Systematic Review: Nicotinamide in Glaucoma—Human Evidence

Last updated:

A systematic review evaluated human evidence on nicotinamide and NAD-targeted strategies in glaucoma. Observational studies reported associations between glaucoma and altered circulating nicotinamide concentrations and systemic mitochondrial dysfunction. The authors consider the question of sustained neuroprotection or modification of glaucoma progression as open.

1. What the Research Says

PubMed, Scopus and the Cochrane Central Register of Controlled Trials were searched through August 2026; interventional, observational and mechanistic studies were included. Available studies were generally small and of limited duration, and the specific contribution of nicotinamide could not be isolated.

2. How It Fits Earlier Research

The new review is consistent with the earlier randomized crossover trial in which nicotinamide improved inner retinal function in glaucoma. Both leave open whether this can also modify the disease course over the long term.
